Growth performance of human adipose-derived mesenchymal stem cells in media containing Simplified Platelet Lysate (SimPL) and fetal bovine serum (FBS). Cells were grown in standard DMEM/F12 basal media containing penicillin, streptomycin, and GlutaMAX™ with either 10% (v/v) FBS or 2% SimPL (left). Similarly, cells were grown in identical conditions except for the use of reduced-serum basal media (Gibco™ Advanced DMEM/F12) with either 2% FBS or 2% SimPL (right). Medias supplemented with SimPL also contained additional magnesium sulfate sufficient to achieve a final media concentration of 1.7 mM Mg. Prior to growth evaluation, cells were passaged using standard techniques in 10% FBS with standard DMEM/F12 basal media and were seeded into each condition without adaptation into a 96-well plate.

Growth performance of 3T3-L1 fibroblasts in media containing Simplified Platelet Lysate (SimPL) and fetal bovine serum (FBS). 3T3-L1 fibroblasts were grown in DMEM/F12 basal media containing penicillin, streptomycin, and GlutaMAX™ with either 10% (v/v) FBS or 10% SimPL. Media supplemented with SimPL also contained additional magnesium sulfate sufficient to achieve a final media concentration of 5.7 mM Mg. 3T3-L1 cells were passaged using standard techniques in each condition 10 times before seeding a 96-well plate for label-free cell counting with a BioSpa 8 automated incubator and Cytation 5 multi-mode reader. Data represent counts of cells present within imaged well center. Comparable performance was seen throughout all prior passages without the need for adaptation to media containing SimPL (data not shown).

Growth performance of Madin-Darby canine kidney (MDCK NBL-2) cells in media containing Simplified Platelet Lysate (SimPL), fetal bovine serum (FBS), or human platelet lysate (hPL). Cells were grown in standard DMEM/F12 basal media containing penicillin, streptomycin, and GlutaMAX™ with either 10% (v/v) FBS, 2.5% SimPL, or 2.5% hPL. Media supplemented with SimPL also contained additional magnesium sulfate sufficient to achieve a final media concentration of 2 mM Mg. Prior to growth evaluation, cells were passaged using standard techniques in 10% FBS with standard DMEM/F12 basal media and were seeded into each condition without adaptation into a 96-well plate. Data represent cell counts relative to the initial seeding density average of each condition obtained from label-free cell counting with a BioSpa 8 automated incubator and Cytation 5 multi-mode reader.

Growth of FO myeloma cells in basal media containing SimPL-Serum. Growth of FO myeloma cells in media containing Simplified Platelet Lysate (SimPL-Serum) and fetal bovine serum (FBS). Cells were grown in standard DMEM basal media containing penicillin, streptomycin, and GlutaMAX™ with 10% (v/v) FBS or 10% SimPL-Serum in 6-well plates. Cells were grown in suspension culture and subcultivated every 3 days at an average cell density of 0.4 x 10^5 cells/mL. Data represent the total number of population doublings since initial cell harvest obtained from total cell counts with a Z Series Beckman Coulter Counter.
