Concanamycin A, also designated folimycin, is an antibiotic and well-noted specific inhibitor of V-ATPase (vacuolar type H+-ATPase) activity. Concanamycin A is also capable of inducing cell-death through apoptosis, able to inhibit perforin-based cytotoxic activity in CD8+ T cells, and to inhibit intracellular protein trafficking in both plant and animal cells. As an inhibitor of V-ATPase, Concanamycin A has been reported to induce cell death via apoptosis in WEHI 231, SP2/0-Ag14 cells lines as well as mouse hybridomas. The inhibition of perforin-based cytotoxic activity in CD8+ CTL clone OE4 was reported to be selective for completely blocking perforin-based cytotoxic activity in these clones. Additionally, concanamycin A was noted to lead to reversible vacuolation of the Golgi apparatus in tobacco BY-2 cells, indicating potential interference with protein trafficking in this cell line.
