IHC-Tek -Model IW-1800 -Rabbit LSAB Detection Kit for Immunohistochemistry
The IHC-Tek Rabbit LSAB Detection Kit is developed for use with user-supplied Rabbit primary antibodies, facilitating the detection of antigens in tissue sections or cell preparations using light microscopy. This kit is particularly suitable for frozen tissue, paraffin-embedded tissue, freshly prepared lymphocytes, fixed culture cells, and also for whole mount IHC staining. A key feature of this detection system is its employment of a horseradish peroxidase (HRP)-labeled streptavidin and biotinylated secondary antibody system, which is widely recognized in immunochemical staining methodology. The enhanced binding strength of the kit's components offers improved sensitivity and reduced non-specific background staining, leading to a higher signal-to-noise ratio in comparison to the traditional avidin-biotin complex (ABC) method. Notably, when using the kit's Antibody Diluent as the primary antibody dilution buffer, the additional step of serum protein blocking may be omitted.
The IHC-Tek Rabbit LSAB Detection kit is intended to use with Rabbit primary antibody (user-supplied) to detect the presence of antigens in tissue sections or cell preparations under light microscopy. Most commonly used specimens for this system are: frozen tissue, paraffin-embedded tissue, freshly prepared lymphocytes and fixed culture cells. It is also suitable for whole mount IHC staining.
Horseradish peroxidase (HRP) labeled-streptavidin and biotinylated secondary antibody amplification system has become a standard technique in immunochemical staining. IHC-Tek LSAB Rabbit Detection kit uses biotinylated, affinity-purified secondary antibody reacts with the user supplied rabbit primary antibody bound to the specific epitope of the antigen in tissue or cell. Horseradish peroxidase (HRP) labeled streptavidin then reacts with biotinylated secondary antibody to form a HRP-streptavidin-biotin complex. The HRP enzyme of the streptavidin complex catalyzed the DAB chromogen substrate (3,3’ diaminobenzidine) reaction to form brown color deposit at the antigen site. The antigen then can be visualized under microscope. Compared to traditional ABC method which uses avidin, IHC-Tek Rabbit LSAB Detection kit demonstrates stronger binding strength to bind biotin and less non-specific background staining.
Higher sensitivity and lower background give the kit a higher signal-noise ratio. When the Antibody Diluent is used as primary antibody dilution buffer, there is no need to perform serum protein blocking step.
