Model Sub-Type 1B -Recombinant Core protein, HCV

SHARE

Recombinant Hepatitis C virus Core protein is expressed in E. coli as a monomeric, non-glycosylated polypeptide chain consisting of 115 amino acids with a molecular mass of 13100 Da. The product represents amino acids 1 to 115 of the HCV polyprotein, covering the N-terminal region (“domain I”) of the mature Core protein. It lacks the C-terminal signal peptide, the transmembrane region and the ER anchoring sequence (“domain II”) of the native protein.

The integrity of the plasmid is controlled by restriction analysis and DNA sequencing. The expression product is characterized by N-terminal protein sequencing, SDS-PAGE (app. m.w.) and immunodetection of the Western blotted protein. In addition, correct reactivities compared to previous antigen lots with defined human and animal sera (negative, low, medium and high positive) are mandatory.
