These disorders stem from abnormalities in three key areas: bone resorption, bone growth, and mineral deposition. Increasingly, research has identified numerous cytokines involved in the processes of bone remodeling and metabolism. Cytokines such as IL-1α, IL-1β, TNF-α, and TNF-β significantly stimulate bone resorption and inhibit bone formation. IL-6, synthesized by osteoblasts (OBs), is promoted by parathyroid hormone (PTH), which encourages OBs to secrete IL-6, thereby facilitating the differentiation and proliferation of osteoclasts (OCs). The role of colony-stimulating factors (CSFs) is critical in the early phases of OC formation and differentiation.
