eSpCas9 (enhanced specificity Cas9) has enhanced specificity compared to wild type Cas9 for CRISPR/Cas9 genome editing. eSpCas9 is able to reduce off-target effects and maintain robust on-target genome editing efficiency, allowing more efficient gene targeting. eSpCas9 (CAG, Neo) lentivirus expresses eSpCas9 under the control of the CAG promoter with neomycin as a selection marker, allowing selecting stable cell clones with neomycin.

CISH (Cytokine Inducible SH2 Containing Protein) contains a SH2 domain and a SOCS box domain. SOCS family proteins form part of a classical negative feedback system that regulates cytokine signal transduction. CISH may be a substrate-recognition component of a SCF-like ECS (Elongin BC-CUL2/5-SOCS-box protein) E3 ubiquitin-protein ligase complex which mediates the ubiquitination and subsequent proteasomal degradation of target proteins.

Lentiviral vector-CAG, BSD contains  Multiple Cloning Sites (MCS) downstream of the CAG promoter, and blasticidin driven by the PGK promoter as a selection marker. Clone your gene of interest into the MCS, then co-transfect with EZ-LentiPACK Packaging System into packaging cells, such as HEK293T to produce VSV-G pseudotyped lentivirus. Transduced stable cells can be selected by blasticidin.
