Description:Lubiprostone (SPI-0211; RU0211) increases intestinal fluid secretion through generation of CIC-2/CFTR and activation of cAMP signaling pathway. Lubiprostone inhibits myeloperoxidase (MPO) activity, downregulates Indomethacin (HY-14397)-induced iNOS and TNFα expression. Lubiprostone can be used for chronic constipation research.
In Vitro:Lubiprostone (250 nM, 20 min-4 h) induces the generation of cAMP and cystic fibrosis transmembrane conductance regulator (CFTR) in T84 cell, increases the secretion of chloride ions[1].
In Vivo:Lubiprostone (0.1-0.5 mg/kg, po, once daily for 5 weeks) ameliorates high fat diet (HFD)-induced non-alcoholic fatty liver disease (NAFLD) in mouse models[2]. Lubiprostone (0.01-1 mg/kg, po, singe dose) reduces Indomethacin (HY-14397)-induced small intestinal damage and prevents enterobacterial invasion in rats models[3].
