Our ALiCE cell-free protein expression kits are designed with ease-of-use in mind. Using our simple protocol will take you from reaction set-up to expressed protein in under 48 hours. All it takes is your gene-of-interest cloned into one of our vectors and the ALiCE®  lysate. No additional reagents, no extra steps, and no protein-specific optimizations.

The ALiCE® cell-free protein expression kits set a new benchmark for protein production. The kits contain a lysate derived from N. tabacum c.v. BY-2 cells which have been specially optimized to produce consistently high protein yields of up to 3 mg per mL (eYFP). The inclusion of native microsomes derived from the endoplasmic reticulum and Golgi apparatus means the lysate is also capable of producing proteins with complex eukaryotic post-translational modifications, including disulfide bonds and glycosylation. The kits are provided with two vectors, pALiCE01, and pALiCE02 respectively, which enable the user to produce proteins via cytosolic or microsomal pathways. These elements combined, provide a powerful expression tool capable of producing even the most difficult-to-express proteins.

Two pathways of expression

The ALiCE® kit offers two routes of protein expression for ultimate flexibility:

  1. For the expression of simple, cytosolic proteins, the pALiCE01 vector is provided. Using this vector, up to 3 mg/ml protein can be achieved.
  2. For expression of more complex eukaryotic proteins, delivery of your gene in the pALiCE02 vector will target your protein to the microsome (derived from Golgi and endoplasmic reticulum), for post-translational modifications.
